PerCP-Cy®5.5 Antibody Conjugates

Leverage the tandem fluorophore PerCP-Cy®5.5 for improved photostability and brightness over PerCP alone. Due to the resonance energy transfer from PerCP to Cy5.5, you can enhance the utility of your 488 laser line, making bright PerCP-Cy®5.5 conjugated antibodies an excellent choice for multiplexed flow cytometry workflows and assays targeting less abundant antigens.

The excitation spectrum (dashed blue line) and emission spectrum (solid blue)  of Alexa Fluor  350 are shown above. Selecting fluorophores with minimal excitation and emission spectral overlap yields the best results when multiplexing. Alexa Fluor  350 has a broad excitation range, allowing it to be excited by ultraviolet (330 nm) and conventional violet lasers (355 nm).

PerCP-Cy®5.5 conjugated antibodies have a far-red emission spectrum (solid red) with reduced emission overlap with many commonly used fluorophores. With an excitation maximum of 482 nm (dashed red line), the same laser line can be used with other fluorophores, such as FITC or PE, for easy integration into multicolor panels.

Applications in Research

Several properties of PerCP-Cy®5.5 make it a powerful research tool, such as:

  • Brighter than PerCP alone due to improved energy transfer. This makes it ideal for detecting low-abundance markers or applications requiring a strong signal strength with limited autofluorescence.
  • Excitable by the standard 488 nm laser, it is a convenient choice, as no additional lasers or modifications to equipment are required.
  • Causes less Fc receptor-mediated nonspecific staining than other standard tandem fluorophore conjugates.
