DyLight 800 Antibody Conjugates

Detect low-abundance proteins and cells with high quantum yield DyLight 800 antibody conjugates. DyLight 800 is a bright, near-infrared dye (800 nm) with a high signal-to-noise ratio and minimal crosstalk with visible spectrum dyes. It is also a photostable dye that is resistant to photobleaching and is compatible with infrared imaging systems.

Western blot analysis of Jurkat cell lysates (#9194) treated with either U0126 (MEK 1/2 inhibitor) #9903 or TPA (12-O-Tetradecanoylphorbol-13-Acetate) #4174, using Phospho-p44/42 MAPK (Erk1/2) (Thr202/204) (D13.14.4E) XP® Rabbit mAb #4370 detected with Anti-rabbit IgG (H+L) (DyLight 800 4X PEG Conjugate) (green) and p44/42 MAPK (Erk1/2) (3A7) Mouse mAb #9107 detected with Anti-mouse IgG (H+L) (DyLight 680 Conjugate) #5470 (red). The array image pixel intensities obtained using a LI-COR Biosciences Odyssey Infrared Imaging System are shown in the panel to the left while corresponding fluorescent western blots are shown in the panel to the right.

DyLight 800 Excitation Spectrum

The dashed line illustrates the excitation spectrum of DyLight 800, while the solid line represents its emission spectrum. In experiments with multiple dyes, best results are obtained by avoiding emission spectral overlap. DyLight 800 has a broad excitation spectrum that peaks around 777 nm but can effectively be excited by a range of NIR wavelengths, from roughly 740 nm to 800 nm.

Applications in Research

Several key advantages make DyLight 800 antibodies powerful tools for researchers:

  • DyLight 800 can be used alongside another near-infrared fluorophore, such as DyLight 680, to perform dual-color Fluorescent Western blotting. This allows researchers to detect and quantify multiple proteins simultaneously on the same blot.
  • It can be used to generate clearer images in thicker samples due to the high tissue penetration of near-infrared light.
  • The high photostability of DyLight 800 antibodies supports accurate quantification and visualization when imaging over extended periods, making it suitable for longitudinal studies in pharmacokinetics and biodistribution.

The photostability of DyLight 800 also makes it applicable to a broad array of experiments, including:

In-Cell Western™ analysis of A549 cells exposed to varying concentrations of U0126 (MEK1/2 Inhibitor) #9903 for 3 hours, followed by TPA (Phorbol-12-Myristate-13-Acetate) #9905 stimulation for 30 minutes. With increasing concentrations of U0126, a significant decrease (~2 fold) in Phospho-p44/42 MAPK (Erk1/2) (Thr202/Tyr204) (E10) Mouse mAb #9106 signal as compared to the TPA-stimulated control was observed. Data and images were generated on the LI-COR Biosciences Odyssey Infrared Imaging System using Anti-mouse IgG (H+L) (DyLight 800 4X PEG Conjugate). DRAQ5® #4084 (fluorescent DNA dye - red) is used for normalization.
