    This Cell Signaling Technology® antibody retains the antigen-binding Fab regions of the original parent host sequence from which it is engineered. This antibody is expected to exhibit the same species cross-reactivity as V5-Tag (D3H8Q) Rabbit mAb #13202.

    Storage

    Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

    Specificity / Sensitivity

    V5-Tag (D3H8Q) Mouse Chimeric mAb detects recombinant proteins containing the V5 epitope tag. The antibody recognizes the V5-tag fused to either the amino or carboxy terminus of targeted proteins in transfected cells. V5-Tag (D3H8Q) Mouse Chimeric mAb detects exogenously expressed V5-tagged proteins in cells expressed under a CMV promoter. Expression under other promoters has not been evaluated.

    Source / Purification

    This recombinant chimeric antibody is engineered from V5-Tag (D3H8Q) Rabbit mAb #13202 according to animal-free protocols. The chimeric antibody retains its antigen-binding Fab regions from the original rabbit monoclonal antibody but contains a mouse-derived Fc domain. When multiplexing, Fc-directed rabbit secondaries are required to detect rabbit-host primary antibodies.

    The parent antibody, V5-Tag (D3H8Q) Rabbit mAb #13202 is produced by immunizing animals with a synthetic V5-tag peptide.

    Background

    Epitope tags are useful for the labeling and detection of proteins using immunoblotting, immunoprecipitation, and immunostaining techniques. Because of their small size, they are unlikely to affect the tagged protein’s biochemical properties.

    The V5 tag is a 14 amino acid peptide derived from a small epitope (Pk) on the P and V proteins of simian virus 5 (SV5), a member of the paramyxovirus family (1). The V5 tag can be used to detect expression of recombinant proteins in bacteria, yeast, insects, and mammalian cell systems.
