    This Cell Signaling Technology® antibody retains the antigen-binding Fab regions of the original parent host sequence from which it is engineered. This antibody is expected to exhibit the same species cross-reactivity as T7-Tag (D9E1X) XP® Rabbit mAb #13246.

    Storage

    Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

    Specificity / Sensitivity

    T7-Tag (D9E1X) XP® Mouse Chimeric mAb detects recombinant proteins containing the T7 epitope tag. This antibody recognizes the T7-tag fused to either the amino or carboxy terminus of targeted proteins in transfected cells. This antibody detects exogenously expressed T7-tagged proteins in cells expressed under a CMV promoter. Expression under other promoters has not been evaluated.

    This recombinant chimeric antibody is engineered from T7-Tag (D9E1X) XP® Rabbit mAb #13246 according to animal-free protocols. The chimeric antibody retains its antigen-binding Fab regions from the original rabbit monoclonal antibody but contains a mouse-derived Fc domain. When multiplexing, Fc-directed rabbit secondaries are required to detect rabbit-host primary antibodies.

    The parent antibody, T7-Tag (D9E1X) XP® Rabbit mAb #13246 is produced by immunizing animals with a synthetic T7-tag peptide.

    Background

    Epitope tags are useful for the labeling and detection of proteins using immunoblotting, immunoprecipitation, and immunostaining techniques. Because of their small size, they are unlikely to affect the tagged protein’s biochemical properties.

    The T7-tag is an epitope tag composed of an 11-residue peptide encoded from the leader sequence of the T7 bacteriophage gene 10. The T7-tag has been used extensively as a general epitope tag in many expression vectors including the pET system that is based on T7 RNA polymerase expression systems (1).
