    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Val50 of human PPP1R14B/PHI-1 protein.

    Background

    Phosphatase holoenzyme inhibitor (PHI-1), encoded by the protein phosphatase 1 (PP1) regulatory subunit 14B gene (PPP1R14B), belongs to a family of proteins, including PPP1R14A/CPI-17, PPP1R14C/KEPI, and PPP1R14D/GEPI that each specifically inhibits a subset of PP1 holoenzymes. PHI-1 is highly conserved among eukaryotes and ubiquitously expressed at low levels across tissue types, but it is overexpressed in a wide array of cancer types, including colon and lung adenocarcinomas (1,2). PHI-1 also displays a proteostatic function, both directly and indirectly, on several targets in the MAPK signaling pathways. In hepatocellular carcinoma (HCC) cells, PHI-1 bound to, stabilized, and increased phosphorylation of p90RSK at Ser363/Ser380, and PHI-1 knockdown inhibited HCC cell growth (3). In HEK 293 cells, PHI-1 knockdown stabilized c-Raf protein and increased downstream ERK1/ERK2 activation (4). PHI-1 phosphorylation at Thr57 increases its inhibitory activity toward PP1 50-fold, and it has been established as a PKC and ILKsubstrate (5,6).
