    Background

    Nectin cell adhesion molecule 3 (Nectin-3), also known as poliovirus receptor-related protein 3 (PVRL3) and CD113, is a member of the immunoglobulin-like cellular adhesion molecule family and plays an important role in calcium-independent cellular adhesion (1,2). There are four nectin family members, nectin-1, nectin-2, nectin-3, and nectin-4. They are composed of an ectodomain containing three tandem Ig-like regions arranged as an N-terminal variable-like domain followed by two constant-like domains, a single transmembrane region, and a cytoplasmic domain that binds to the actin cytoskeleton through the adaptor protein afadin (3,4). Nectin-3 can engage in homophilic, as well as heterophilic cell adhesion by engaging with other nectin family members (5). In the nervous system, nectin-3 expression is associated with chronic stress-induced synaptic abnormalities and has been proposed as a target for stress-related cognitive disorder treatment (6,7). Nectin-3 can also interact with the immune system. Nectin-3 expressed on antigen-presenting cells (APCs) and tumor cells can act as a ligand for the inhibitory receptor TIGIT, expressed on activated T cell subsets and natural killer (NK) cells (8). Aberrant expression of nectin-3 has been observed in multiple cancers, including breast, pancreatic, lung, colorectal, and ovarian cancer (9-14).
