    Background

    Nectin cell adhesion molecule 1 (Nectin-1), also known as poliovirus receptor-related 1 (PVRL1) and CD111, is a single-pass type I membrane glycoprotein and is part of the immunoglobulin (Ig) superfamily of cell adhesion proteins (1). Nectins are composed of an ectodomain containing three tandem Ig-like regions arranged as an N-terminal variable-like (V) domain followed by two constant-like (C) domains, a single transmembrane region, and a cytoplasmic domain that binds to the actin cytoskeleton through the adaptor protein afadin (2). Nectin-1 plays important roles in adherens junction (AJ) formation in epithelial cells and neuronal synapses (1,3). Like other nectin family members, nectin-1 is used as an entry receptor for several mammalian viruses, including herpes simplex virus types 1 and 2 (HSV-1 and HSV-2), porcine pseudorabies virus (PRV), bovine herpes virus type 1 (BHV-1), and simian B virus (4-6). Nectin-1 also plays a role in the immune system, where it interacts with CD96 expressed on natural killer (NK) cells and T cell subsets, and modulates cancer cell and virally infected cell clearance (7,8).
