    Protocol

    Specificity / Sensitivity

BARD1 (F4K3V) Rabbit Monoclonal Antibody recognizes endogenous levels of total BARD1 protein. Based on the sequence of the protein antigen, this antibody is expected to detect the alpha and beta isoforms of BARD1.

    Species Reactivity:

    Human

    Source / Purification

Monoclonal antibody is produced by immunizing animals with recombinant protein specific to the amino terminus of human BARD1 protein. This antibody recognizes residues surrounding Arg155 of human BARD1 protein.

    Background

BRCA1-associated RING domain protein 1 (BARD1) is an E3 ubiquitin-protein ligase that acts as a tumor suppressor as part of a BRCA1-BARD1 heterodimer. This complex plays a crucial role in maintaining genomic stability by regulating various cellular processes, including DNA repair, cell cycle checkpoint control, and transcription, through its E3 ubiquitin ligase activity (1). The BRCA1/BARD1 heterodimer is required for DNA end resection during repair of DNA double-strand breaks (DSBs) through homologous recombination (HR), and protects DNA during replication stress (2-4). BARD1 also functions independently of BRCA1, including in the regulation of p53-dependent apoptosis (5). Several isoforms arising from alternative splicing of the BARD1 gene are differentially expressed in cancer. In addition, mutations and deletions are associated with various cancer types, including neuroblastoma, lung, breast, and cervical cancers (6). BARD1 is a potential therapeutic target in glioblastoma and pancreatic ductal adenocarcinoma (PDAC) (7,8).
